چکیده

One of the few naturally occurring acetyl derivatives in the animal body is that of glucosamine. It is well known that glucosamine is present in a large number of polysaccharides in many organisms and almost always is present there in acetylated form. During studies in this laboratory on the acetylation reaction, we, therefore, became interested in the possibility of carrying out an enzymatic acet.ylation of glucosamine in vitro. Pigeon liver extract had previously been used extensively in experiments on enzymatic acetylation and had been found to contain acetylating systems for several amines (1, 2). It will be reported here that this extract likewise contains the system for acetylation of glucosamine. As with previously studied acet, ylat.ion reactions, glucosamine acetylation is also dependent on coenzyme A (CoA) or, more precisely, acetylwCoA is the immediate acetyl donor in this reaction.

Influence of Certain Cations on Activity of Acetic Thiokinase from Spinach Leaves.

The first evidence for a cofactor necessary in acetate metabolism was provided by Nachmansohn and Machado (13) who discovered a soluble enzyme system in extracts of brain tissue which catalyzed the acetylation of choline in the presence of adenosine triphosphate (ATP). Lipmann (8) and Kaplan and Lipmann (5) described an enzyme system in extracts of pigeon liver which catalyzed the ATP and coenz...

The acetate activating enzyme system of heart muscle.

Nachmansohn and Machado (1) discovered a soluble enzyme system in extracts of brain tissue which catalyzed the formation of active acetate in the presence of ATPI and CoA. Active acetate was detected by the acetylation of choline. Later Lipmann (2, 3) reported an enzyme system in extracts of pigeon liver which catalyzed the CoA and ATP-dependent acetylation of sulfanilamide by acetate. The iden...

N-Acetylation of glucosamine-6-phosphate in Leuconostoc mesenteroides.

A partially purified enzyme (120-fold) from Leuconostoc mesenteroides catalyzed the reversible N-acetylation of d-glucosamine-6-phosphate. Coenzyme A was not required and inhibited the reaction rate. Neither d-glucosamine nor N-acetyl-d-glucosamine served as a substrate for the reversible reaction. The enzyme preparation retained 50% of its original activity after 5 min at 100 C. The K(m) for a...

The enzymatic acetylation of amines.

The enzymatic acetylation of aromatic amines (1, Z), choline (3), glucosamine (4), and histamine (5) has been shown to require coenzyme A (CoA) ; recent studies (6) have demonstrated that the thio ester, acetyl CoA, can serve as the acetyl donor in the acetylation reaction. However, more detailed studies on the properties of the acetylating enzyme were made difficult by the absence of a suitabl...
